Active transport of potassium by the cecropia midgut. VI. Microelectrode potential profile.

چکیده
Potential differences can only appear across barriers with high electrical resistance. Therefore it should be possible to locate the potential produced by an electrogenic pump across such high resistance barriers by using intracellular microelectrodes. An opportunity to locate such a pump is afforded by the electrogenic active transport of K across the midgut of Hyalophora cecropia (Harvey, Haskell & Nedergaard, 1968). The structure of the midgut encourages the attempt to locate the electrogenic pump. Two types of cell, columnar cells (65 /i tall) and goblet cells (35 fi tall) are arranged in a single layer one cell thick to make up the epithelium. Its basal surface is covered by a basement lamina (10 ft thick), surrounded by an inner sheet of circular muscles and outer bundles of longitudinal muscles, and supplied with tracheoles (Anderson & Harvey, 1966). Presumably the midgut potential is generated by the epithelial cells because they form the only continuous cellular layer in the midgut. The first step in locating the potassium pump is to determine where within the epithelial cells the electrogenic potential appears.

Active transport by the cecropia midgut. V. Loss of potassium transport during larval-pupal transformation.
Few serious attempts have been made to study either alterations of active ion transport during ontogenic development or the developmental consequences of altered ion environments within and around cells (see Kroeger, 1966 and Harvey & Haskell, 1966 for references). Nevertheless, to cite but a few examples of the importance of the ionic mileau for molecular activities associated with development...
متن کاملActive transport of sodium by the isolated midgut of Hyalophora cecropia.
Potassium is actively transported from blood-side to lumen of the isolated silkworm midgut (Harvey & Nedergaard, 1964). Rubidium is also actively transported and, when present in equal amounts, potassium and rubidium compete on an almost equal basis (Nedergaard & Harvey, 1968). Recently, Zerahn (1970) has found that caesium as well is actively transported, but the competition with potassium is ...
متن کاملActive transport of potassium by the Cecropia midgut; tracer kinetic theory and transport pool size.
1. Tracer influx kinetics have been analysed theoretically to determine the size of the transport pool with no assumptions regarding the transport pathway. 2. For a calculation of the size of the transport pool to be made, the following six conditions are required by the theory: tracer steady state attained, tissue steady state attained, Isc measures next flux, small magnitude and constant time...
متن کاملActive transport of calcium across the isolated midgut of Hyalophora cecropia.
1. The net flux of 45Ca from lumen to blood side across the isolated and short-circuited Cecropia midgut was 1-9 +/- 0-2 muequiv. cm-2h-1 in 8 mM Ca and the flux ratio was as high as 56 to 1. 2. The calcium influx was depressed by anoxia; 73% after 30 min. 3. The kinetics of Ca transport were anomalous; the apparent Km varied with Ca concentration from less than 0-2 to greater than 5-6 mM Ca an...
متن کاملActive transport of potassium and oxygen consumption in the isolated midgut of Hyalophora cecropia.
Oxygen uptake is essential for active ion transport across the isolated silkworm midgut as well as across other isolated epithelial membranes such as the frog skin and toad bladder. The study of the chemical relationship between these two processes in the skin and bladder has been rewarding. The short-circuit current across the isolated frog skin and toad bladder is accounted for entirely by th...
